Yes, BD confirms that this will work for trucks equipped with a manual transmission. The brake will work just as effectively on manual transmission trucks as it does on automatics. The only difference is you disregard the connection we make with the Tow/Haul button, and the brake will have only one level (level 2 just downshifts for you, so of course you do not need that).
